WP 1 - DESIGN

  1. DESIGN will be devoted to an analysis and optimization of the properties of the building blocks (biopolymers and aptamers) and their ability for the formation of stable delivery platforms that will protect and deliver the payloads to their targets. Furthermore, WP 1 will be focused on the development and optimization of the biopolymer/aptamer complexation and preparation of core-shell capsules, and their additional functionalization through co-encapsulation of potential therapeutic agents.

WP 2 - CHARACTERIZATION

  1. CHARACTERIZATION will be addressed to investigation of the properties (surface characteristics, internal morphology, stability at physiological conditions, and encapsulation efficiency of the drugs) of the produced aptamer-drug delivery systems.

WP 3 - FUNDAMENTAL

  1. FUNDAMENTAL will be focused on investigation of the interactions between selected aptamer loaded capsules (from WP 1 and WP 2) and spike glycoproteins on the model biological membranes (lipid layers). The results of WP 3 will continuously provide feedback to the optimization strategies in WP 1.

WP 4 - IN VITRO antiviral activity assay

  1. IN VITRO antiviral activity assay will be devoted on investigation of the cytotoxicity and anti-coronavirus activity of the produced capsules against MRC-5 cell lines.

WP 5 - IN VITRO virus attachment assay

  1. IN VITRO virus attachment assay will be focused on the analysis of the interaction between selected aptamer loaded capsules and spike protein of HCV/229E. The assays will estimate the effectiveness of the blockage of the viral particle even before contact with sensitive cells, as well as interacting with the cell membrane to make it impermeable to the virus and the viral invasion to be stopped in early stage. The results of WP 5 will provide complementary information and feedback to WP 3.

WP 6 - IN VITRO oxidative stress assay

  1. IN VITRO oxidative stress assay will have for an object to determine the degree of oxidative stress in diploid cell culture MPC-5 in the presence of loaded capsules at a state of coronavirus infection.

WP 7 - INNOVATION MANAGEMENT

  1. INNOVATION MANAGEMENT will be addressed to dissemination of the results of the project.

WP 8 - MANAGEMENT

  1. MANAGEMENT will take over all legal, financial, and administrative aspects of project managing.
